Abstract
To test how methylation affects protein function in translation, we used two Ded1p methyl mutants, whose methylated arginine residues have been substituted so that the protein cannot be methylated. After phenotyping the mutants, we did not detect any global translation defect. We therefore moved to testing specific transcripts to look for a difference in translation in the methyl mutants. Using in vivo luciferase assays, we measured the translation efficiency of specific mRNAs that are known to have long and highly structured 5' UTRs. We have found that one of these mRNAs has a significantly lower translation efficiency in Ded1p methyl mutant cells.
